چکیده

Based on their observations that the lipoxidase-catalyzed oxidation of linoleate, like autoxidation, gave primarily conjugated diene peroxides (1,2) and caused a coupled oxidation of some easily oxidizable substances, such as carotenoids (3) and antioxidants (4), Bergstrom and Holman proposed a reaction mechanism for lipoxidase catalysis. They suggested (4) that lipoxidase could cause initiation of a free radical chain reaction similar to that operative in the autoxidation of methyl linoleate (5). Because the lipoxidase reaction yields peroxides whose molecular extinction coefficient is considerably greater than the molecular extinction coefficient of peroxides produced in autoxidation (4) under the same conditions, Lundberg (6) suggested that lipoxidase may not function merely to initiate a chain reaction like that prevailing in autoxidation but may form a complex with each linoleate molecule and thus influence the structure of the resulting peroxides. More recently Kunkel (7) reported results that were interpreted as being in accord with a chain reaction mechanism for lipoxidase catalysis. He found that the rate of oxidation of methyl linoleate measured by bixin decolorization was proportional to the square root of the chain initiator (lipoxidase) concentration and, therefore, conformed to the same kinetic relationships that had previously been reported for the autoxidation of ethyl linoleate (5). Direct proportionality between reaction velocity and the square root of the enzyme concentration would in general be expected if a chain mechanism is operative (8). Kunkel also reported data on the inhibition of lipoxidase by CYand r-tocopherols, from which he calculated that the minimum chain length in his system was 12 molecules. An understanding of lipoxidase catalysis is of interest in relation to proposals that some enzyme reactions may proceed by chain reactions (g-12). Other than for lipoxidase, speculations on possible chain mechanisms are largely without experimental support. It was the purpose of

Induction period in the lipoxidase-catalyzed oxidation of linoleic acid and its abolition by substrate peroxide.

The non-enzymatic oxidation of linoleate occurs with a definite induction period. Bolland has explained the kinetics of the oxidation in terms of a chain reaction involving free radicals (1). The discovery by Privett et al. (2) that the peroxide formed by the action of lipoxidase is optically active clearly rules out the possibility of a non-enzymatic chain sequence, but it is not impossible th...

Rumen Degradability and Model Prediction of Nutrient Supply to Ruminants from Different Processed Soybean Meals

DVE2010 system was used for model prediction of nutrient supply to ruminants from soy bean meal (SBM), extruded soy bean meal (SBE) and full-fat soybean meal (FSBM). Extruded soy bean meal had the highest truly absorbed rumen undegraded protein in the small intestine (ARUP) followed by SBM and FSBM. There was no significant difference between SBE and FSBM in the case of truly absorbed rumen syn...

Chemotropic Response of the Pollen of Antirrhinum majus to Calcium.

An improved spectrophotometric method for the determination of lipoxidase activity was developed and applied in studies of the purified enzyme and crude enzyme preparations from leguminous seeds, with linoleic acid solubilized in Tween 20 as the substrate. The optimum pH was found to be 7.0, 6.5, 6.0, and 5.5, for purified soybean lipoxidase and for the crude lipoxidases extracted from gram flo...

Evaluation of bean and soy tempeh influence on intestinal bacteria and estimation of antibacterial properties of bean tempeh.

In this study the effect of bean tempeh on the growth of Bacillus subtilis, Escherichia coli, Lactobacillus acidophilus and Lactobacillus paracasei bacteria was investigated. Antibacterial activity was observed only in relation to the bacteria Bacillus subtilis. The effect of tempeh products on human intestinal microflora was also assessed. Bean and soy tempeh were culinarily processed and next...

Biliary lipid composition and gallstone formation in rabbits fed on soy protein, cholesterol, casein and modified casein.

In four experimental groups, rabbits were fed on diets containing soy beans, soy beans plus cholesterol (1%, w/w), casein and modified casein for 8 weeks. Biliary lipid levels, lithogenic-index values and the rate of gallstone formation were determined. The highest mean relative concentrations (mol%) of cholesterol and phospholipid were found in the soy bean + cholesterol group, and the highest...
